Practical Application

 

Tacrolimus, also known as FK506, was first isolated from Streptomyces tsukubaensis by the Fujisawa Pharmaceutical Industrial Research Institute in Japan in 1984. Its chemical structure is a 23-membered macrocyclic lactone antibiotic. It is a potent new immunosuppressant that primarily works by inhibiting the release of interleukin-2, effectively suppressing the action of T lymphocytes, being 100 times more potent than cyclosporine.

On June 7, 1993, tacrolimus was first approved for marketing in Japan, followed by several dozen countries including the UK, the US, and Germany. It is used for acute anti-rejection therapy in liver, kidney, and bone marrow transplants. Formulations developed include capsules, injections, and ointments. Due to the crystalline structure of raw tacrolimus materials, the preparation of a solid dispersion is aimed at rendering it amorphous. Current technology is limited to preparing solid dispersions of tacrolimus, but if formulated to an amorphous form using formulation technology, the same effect can be achieved, with no aging issues associated with solid dispersions. Dissolving tacrolimus in ethanol and using this solution as a binder to granulate excipients results in rapidly dissolving tacrolimus capsules, although the viscosity of tacrolimus ethanol solution is low, leading to poor flowability of the granules and significant differences in dosing volume.

The inventors jointly employed cyclodextrin complexation technology, adding hydroxypropyl β-cyclodextrin, to prepare an amorphous tacrolimus complex through simple granulation and drying, dissolving hydroxypropyl β-cyclodextrin in ethanol with a certain viscosity for use as a binder, resulting in granules with good flowability.
